Urgent Help Needed‼️
*Refugee Children with Cancer Appeal*
The REF team are undertaking to support two young refugee children diagnosed with cancer, both of whom are being cared for by the paediatric oncology team at PPUKM, one of the only hospitals that are able to treat them. We have been told that both children have a good chance of survival with the respective treatments prescribed. The children and their individual stories are as follows:
1️⃣ Somina Begum is a 4-year-old Rohingya girl diagnosed with stage 4 Wilm's tumor, a rare form of kidney cancer in children. She is the youngest of two siblings, her parents and older sibling having sought refuge in Malaysia several years before she was born. Her family used to reside in JB but have moved to KL in recent weeks to begin her first set of chemo sessions. She will then undergo a surgery to remove her affected kidney, followed by further chemo & radiotherapy sessions. The total estimated cost for Somina is *RM100k*.
2️⃣ Joshua Mang is a 10-month-old baby boy from Myanmar, diagnosed with neuroblastoma. He is the only child to a couple who moved to Malaysia 6 years ago. Joshua’s father works as a waiter while his mother is a housewife. Joshua will also be undergoing chemo treatment and surgery, with an estimated total cost of about *RM50k*.
Refugee Emergency Fund - REF is pledging to allocate RM60k of our reserved funds to support both Somina and Joshua (RM15k has been spent). We therefore urgently need your help with further funds of RM90k so that these young ones are able to complete their treatment and get back to health.

【大數據探勘發現老藥合併治療新策略】
生命科學系暨生醫電子與資訊學研究所阮雪芬教授與國立陽明大學生物醫學資訊所黃宣誠教授的合作研究團隊，在國家衛生研究院和科技部資助下，運用美國國家衛生研究院所資助LINCS計劃（Library of Integrated Network-based Cellular Signature）藥物擾動基因表現圖譜公開巨量資料，開發一系列計算生物與系統生物學分析方法，成功地找到具有新用潛力之已上市藥以及新穎癌症組合藥物治療。上述相關研究成果陸續發表於美國癌症研究學會出版的權威期刊《Clinical Cancer Research》和Cell子期刊《iScience》。Cell與Science和Nature並列為世界三大頂級期刊之一。主要研究工作由博士生黃振綜（第一作者）和謝巧慧（第二作者）完成。參與研究的團隊成員還包括臺大醫院許文明醫師團隊和生醫電子與資訊學研究所歐陽彥正教授。
相較於傳統藥物開發環繞在「單一藥物結合單一標靶」之概念，近年來藥物實際上普遍具有之「多標靶藥理」特性（即單一藥物能結合多重標靶）逐漸受到醫學界重視，甚至對於某些疾病之有效治療是必需的。本研究團隊利用LINCS巨量藥物擾動基因圖譜資料（gene expression profiling），剖析複雜多標靶藥理現象，藉以探索舊藥新用與組合治療之可能性，縮短藥物研發總時程。
首先我們利用藥物擾動基因表現變化強度大小之特點，開發一個衡量藥物之間相似度的新度量（metric），並證明這個新度量比其他常用度量能更準確地將已知相同作用機制之藥物分類在一起，成功運用它找到可能具有拓樸酶抑制劑活性的非抗癌用上市藥。
癌細胞生長除了仰賴自身致癌突變基因，也會高度依靠一些非突變基因之功能，這兩種現象分別稱為「癌基因成癮」（oncogene addiction）與「非癌基因成癮」（non-oncogene addiction）。我們藉由分析LINCS巨量資料，系統性揭示藥物誘發基因標記（gene-expression signatures），發現其與癌症特徵（cancer hallmark）以及藥物敏感性（drug sensitivity）息息相關，並進一步完成相關藥效評估試驗。本研究從非癌基因成癮之角度，提供新穎藥物與組合治療策略，在精準醫學（precision medicine）上具有潛在應用價值。
神經母細胞瘤（neuroblastoma）是一種罕見兒童惡性腫瘤，其基因突變之高度異質性，使得標靶治療應用仍相當有限。此外高危險群患者其五年存活率僅不到40%。為尋找有效治療藥物，我們利用GEO公開資料庫（Gene Expression Omnibus），針對近千名神經母細胞瘤患者腫瘤基因圖譜資料進行整合分析，發現與高危險群相關之非癌成癮基因，並結合LINCS藥物誘發基因標記成功預測了一些具有潛力之新藥，並在老鼠體內驗證抗寄生蟲藥「耐克螺」（niclosamide）之抗神經母細胞瘤活性，並利用蛋白質體學闡述其作用機制。
