《俠盜獵車手V》是一款第三人稱開放世界動作冒險遊戲。玩者可通過完成帶設定目標的任務場景，使故事取得進展。除了做任務，玩家可自由徜徉開放世界裡--從《聖安地列斯》中的開闊鄉村地區，到虛構城市洛聖都。《俠盜獵車手V》的地圖比系列的任一早期作品都大。地圖從一開始就可不受限制地探索，但隨著劇情的發展，會有更多的遊戲內容解鎖。玩者使用拳腳近身攻擊、槍械、武器和炸彈等來與敵人戰鬥，並能夠以跑步、跳躍、游泳和駕駛各種載具等方式瀏覽地圖。
